EGCG类脂质体及其冷冻解冻凝胶的制备与表征

Preparation and characterization of the EGCG-loaded niosome and its cryogel

摘要:

以吐温60、胆固醇为壁材,通过乙醇注入法制备了EGCG类脂质体(EN),研究了水化方法及水化介质对EN粒径、PDI、包封率及载量的影响。结果显示,当选取去离子水为水化介质,以12 000 r/min的速率高速分散1 min时制得的EN粒径较小,为61.4 nm,PDI为0.189±0.020;包封率及载量较高,分别为79.4%和7.97%。采用冷冻解冻法将EN嵌入到PVA/HA冷冻解冻凝胶中制得PVA/HA/EN冷冻解冻凝胶。通过SEM表征发现EN的载入使凝胶产生孔状结构,且EN没有被破坏,保持原有粒径大小。通过对PVA/HA/EN冷冻解冻凝胶释放率的研究,发现当EGCG浓度相同时,PVA/HA/EN冷冻解冻凝胶比直接运载EGCG的PVA/HA/ES冷冻解冻凝胶具有更好的缓释效果。

Abstract:

EGCG-loaded niosomes(EN)were prepared by ethanol injection method using Tween 60 and cholesterol as encapsulation materials. The effects of hydration methods and hydration media on the particle size, PDI, entrapment efficiency and loading capacity of EN were studied. The results showed that, when deionized water was used as hydration medium, the EN prepared at 12 000 r/min for 1 min had small particle size of 61.4 nm and a PDI of 0.189±0.020. High entrapment efficiency and loading capacity were achieved as 79.4% and 7.97%, respectively. The PVA/HA/EN cryogels were prepared by embedding EN into PVA/HA cryogels by freeze-thaw method. SEM characterization showed that the presence of EN in PVA/HA cryogels resulted in a porous structure, and EN was not damaged and kept the original particle size. Through the study on the release rate of PVA/HA/EN cryogels, it was found that PVA/HA/EN cryogels had better sustained release effects than the PVA/HA/ES cryogels which directly loaded with EGCG, when the EGCG concentration was the same.
