EGCG阳离子醇质体的制备及性能评价

摘要:

采用乙醇注入法,同时引入阳离子表面活性剂双十八烷基二甲基氯化铵(TA-100),制备了一种阳离子醇质体,并对其制备条件进行了优化。通过透射电镜和动态光散射技术对阳离子醇质体的形貌、粒径分布及Zeta电位等进行了表征。以表没食子儿茶素没食子酸酯(EGCG)为亲水性模型活性物质,探究了制备的阳离子醇质体对EGCG的包载性能,并对其体外释放特性及储存稳定性进行了研究。结果表明,醇质体中阳离子表面活性剂TA-100的引入不仅可有效提高醇质体自身的稳定性,而且可明显提升其对EGCG的包载能力。该阳离子醇质体稳定性良好,对活性物质的包载能力强,缓释性能优异,在医药、化妆品等领域具有广阔的应用前景。

Abstract:

The cationic ethosomes were prepared by ethanol injection method which were added with the cationic surfactant dioctadecyl dimethyl ammonium chloride (TA-100). The preparation conditions were optimized. The morphology, size distribution and Zeta potential of cationic ethosomes were determined by transmission electron microscopy and dynamic light scattering. Using epigallocatechin gallate (EGCG) as the hydrophilic model active substance, the encapsulation efficiency and in vitro release of EGCG loaded cationic ethosomes prepared with TA-100 were investigated, and their storage stability was also characterized. The results showed that the introduction of the cationic surfactant in ethosomes could not only effectively improve the stability of the ethosomes, but also significantly enhance the ability to encapsulate EGCG. The cationic ethosomes had good stability, high encapsulation capacity for active substances, and excellent slow release property, which showed good prospect for application in medicine, cosmetics and other fields.
