Study on the repairing effect of Dong’ e Ejiao on skin barrier injury in mice

Abstract:

The effect of Dong’e Ejiao on repairing mechanical skin barrier injury in mice was investigated. A mouse skin barrier injury model was established using the tape-stripping method. Transepidermal water loss(TEWL)at the damaged skin site was measured after model establishment. The repair effect of donkey-hide gelatin on skin barrier damage was observed through the C-Cube multi-functional skin imaging instrument. Optical coherence tomography(OCT)imaging and HE staining were used to observe the effect of donkey-hide gelatin on the thickness of injured skin. The mRNA levels of filaggrin(FLG), involucrin(IVL), tight junction protein zonula occludens-1(ZO-1), and inflammatory factors interleukin-1α(IL-1α)and interleukin-1β(IL-1β)were detected by RT-qPCR. The protein levels of skin barrier-related proteins were examined by immunohistochemistry(IHC), and its regulatory effect on thymic stromal lymphopoietin(TSLP)was verified by immunofluorescence(IF). The results show that Dong’e Ejiao significantly reduces TEWL in tape-stripped mice, decreases skin thickness, improves disordered epidermal structure, up-regulates the expression of FLG, IVL, and ZO-1, and suppresses the expression of TSLP and down-regulates the mRNA levels of inflammatory factors including IL-1α and IL-1β. These results indicate that Dong’e Ejiao repairs the skin barrier by modulating the expression of barrier proteins and inflammatory factors. This demonstrates its potential for application in the field of skin health and provides a theoretical basis for such use.
