紫外可见光谱分析海娜粉中的指甲花醌

UV-vis spectral analysis of lawsone in henna powder

摘要: 通过分析海娜粉浸提液和不同pH介质中指甲花醌溶液的紫外可见光谱,研究了分光光度法定量测定海娜粉中指甲花醌含量的检测波长和参比溶液。选择453 nm作为检测波长,以pH=2盐酸介质中的海娜粉浸提液作为参比,测定pH=12的氢氧化钠介质中试液的吸光度值,根据标准曲线法定量分析海娜粉中的指甲花醌。结果表明,指甲花醌质量浓度在10~140 mg/L内符合朗伯比尔定律,线性相关系数r=0.999 95,精密度RSD不高于0.47%。样品溶液吸光度值在14 h内无明显变化,指甲花醌加标回收率为99.0%~100.0%。本实验方法操作简便,结果可靠,可作为海娜粉中指甲花醌含量检测的依据。

Abstract: UV-vis spectra of henna aqueous extract and lawsone solution of different pH mediums were analyzed to select the detection wavelength and the reference solution for determining lawsone content in henna powder by spectrophotometer.Selecting 453 nm as the detection wavelength,with henna powder extract in hydrochloric acid of pH value 2 as the reference solution,the absorbance value of the testing solution samples in NaOH solution of pH=12 as the working medium,the lawsone mass concentration in the henna powder sample was quantified based upon comparison with the standard calibration curve.The results showed that within the lawsone mass concentration range of 10~140 mg/L,the absorbance follows Lambert-Beers law;the linear correlation coefficient is r=0.999 95 and the precision RSD is less than 0.47%.The absorbancevalue of the sample solution can keep stable within 14 h,while the lawsone spiked recoveries achieves 99.0%~100.0%.The method is simple,reliable,and can be used to detect the lawsone content in henna powder.
